NM_001378454.1(ALMS1):c.4886G>C (p.Arg1629Pro) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R1630P variant (also known as c.4889G>C), located in coding exon 8 of the ALMS1 gene, results from a G to C substitution at nucleotide position 4889. The arginine at codon 1630 is replaced by proline, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
