Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006361.6(HOXB13):c.705dup (p.Ala236fs), citing Ambry Variant Classification Scheme 2023: The c.705dupT variant, located in coding exon 2 of the HOXB13 gene, results from a duplication of T at nucleotide position 705, causing a translational frameshift with a predicted alternate stop codon (p.A236Cfs*3). This alteration is expected to result in protein truncation or nonsense-mediated mRNA decay. However, loss of function of HOXB13 has not been established as a mechanism of disease. Based on the available evidence, the clinical significance of this alteration remains unclear.
