NM_006361.6(HOXB13):c.391C>A (p.Pro131Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P131T variant (also known as c.391C>A), located in coding exon 1 of the HOXB13 gene, results from a C to A substitution at nucleotide position 391. The proline at codon 131 is replaced by threonine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
