Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.1871C>G (p.Ala624Gly), citing Ambry Variant Classification Scheme 2023: The p.A624G variant (also known as c.1871C>G), located in coding exon 10 of the ALK gene, results from a C to G substitution at nucleotide position 1871. The alanine at codon 624 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
