Uncertain significance — the classification assigned by Ambry Genetics to NM_001130009.3(GEN1):c.97C>G (p.Leu33Val), citing Ambry Variant Classification Scheme 2023: The p.L33V variant (also known as c.97C>G), located in coding exon 1 of the GEN1 gene, results from a C to G substitution at nucleotide position 97. The leucine at codon 33 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
