Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001382430.1(AKT1):c.323A>T (p.Asp108Val), citing Ambry Variant Classification Scheme 2023: The p.D108V variant (also known as c.323A>T), located in coding exon 4 of the AKT1 gene, results from an A to T substitution at nucleotide position 323. The aspartic acid at codon 108 is replaced by valine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
