NM_000152.5(GAA):c.1869G>C (p.Gln623His) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q623H variant (also known as c.1869G>C), located in coding exon 12 of the GAA gene, results from a G to C substitution at nucleotide position 1869. The glutamine at codon 623 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
