NM_001369.3(DNAH5):c.7091G>C (p.Gly2364Ala) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G2364A variant (also known as c.7091G>C), located in coding exon 43 of the DNAH5 gene, results from a G to C substitution at nucleotide position 7091. The glycine at codon 2364 is replaced by alanine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
