Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_020207.7(ERCC6L2):c.1458C>G (p.Phe486Leu), citing Ambry Variant Classification Scheme 2023: The p.F486L variant (also known as c.1458C>G), located in coding exon 9 of the ERCC6L2 gene, results from a C to G substitution at nucleotide position 1458. The phenylalanine at codon 486 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
