NM_001430.5(EPAS1):c.853G>T (p.Asp285Tyr) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D285Y variant (also known as c.853G>T), located in coding exon 7 of the EPAS1 gene, results from a G to T substitution at nucleotide position 853. The aspartic acid at codon 285 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
