Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_016938.5(EFEMP2):c.60G>T (p.Leu20Phe), citing Ambry Variant Classification Scheme 2023: The p.L20F variant (also known as c.60G>T), located in coding exon 1 of the EFEMP2 gene, results from a G to T substitution at nucleotide position 60. The leucine at codon 20 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
