Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_016938.5(EFEMP2):c.719C>T (p.Ser240Phe), citing Ambry Variant Classification Scheme 2023: The p.S240F variant (also known as c.719C>T), located in coding exon 6 of the EFEMP2 gene, results from a C to T substitution at nucleotide position 719. The serine at codon 240 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
