Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_018139.3(DNAAF2):c.1596A>C (p.Glu532Asp), citing Ambry Variant Classification Scheme 2023: The p.E532D variant (also known as c.1596A>C), located in coding exon 1 of the DNAAF2 gene, results from an A to C substitution at nucleotide position 1596. The glutamic acid at codon 532 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
