Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.1774A>C (p.Lys592Gln), citing Ambry Variant Classification Scheme 2023: The p.K592Q variant (also known as c.1774A>C), located in coding exon 10 of the DICER1 gene, results from an A to C substitution at nucleotide position 1774. The lysine at codon 592 is replaced by glutamine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
