NM_177438.3(DICER1):c.2347G>A (p.Asp783Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D783N variant (also known as c.2347G>A), located in coding exon 14 of the DICER1 gene, results from a G to A substitution at nucleotide position 2347. The aspartic acid at codon 783 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
