NM_177438.3(DICER1):c.5434T>C (p.Phe1812Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F1812L variant (also known as c.5434T>C), located in coding exon 24 of the DICER1 gene, results from a T to C substitution at nucleotide position 5434. The phenylalanine at codon 1812 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
