NM_177438.3(DICER1):c.2452G>A (p.Val818Met) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V818M variant (also known as c.2452G>A), located in coding exon 15 of the DICER1 gene, results from a G to A substitution at nucleotide position 2452. The valine at codon 818 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
