NM_001903.5(CTNNA1):c.419T>G (p.Ile140Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I140S variant (also known as c.419T>G), located in coding exon 3 of the CTNNA1 gene, results from a T to G substitution at nucleotide position 419. The isoleucine at codon 140 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
