Uncertain significance for Hereditary pancreatitis — the classification assigned by Ambry Genetics to NM_001868.4(CPA1):c.298GAGCAG[3] (p.Gln103_Met104insGluGln), citing Ambry Variant Classification Scheme 2023: The c.304_309dupGAGCAG variant (also known as p.E102_Q103dup), located in coding exon 3 of the CPA1 gene, results from an in-frame duplication of GAGCAG at nucleotide positions 304 to 309. This results in the duplication of 2 extra residues (EQ) between codons 102 and 103. This amino acid region is not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear.
