Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000089.4(COL1A2):c.3530ACT[3] (p.Tyr1178_Trp1179insTyr), citing Ambry Variant Classification Scheme 2023: The c.3533_3535dupACT variant (also known as p.Y1178dup), located in coding exon 50 of the COL1A2 gene, results from an in-frame duplication of ACT at nucleotide positions 3533 to 3535. This results in the duplication of an extra tyrosine residue between codons 1178 and 1179. This amino acid position is highly conserved in available vertebrate species. Based on the available evidence, the clinical significance of this variant remains unclear.
