NM_000075.4(CDK4):c.65A>C (p.Lys22Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K22T variant (also known as c.65A>C), located in coding exon 1 of the CDK4 gene, results from an A to C substitution at nucleotide position 65. The lysine at codon 22 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
