Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000075.4(CDK4):c.671G>A (p.Gly224Asp), citing Ambry Variant Classification Scheme 2023: The p.G224D variant (also known as c.671G>A), located in coding exon 5 of the CDK4 gene, results from a G to A substitution at nucleotide position 671. The glycine at codon 224 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
