NM_016507.4(CDK12):c.4302G>C (p.Glu1434Asp) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1434D variant (also known as c.4302G>C), located in coding exon 14 of the CDK12 gene, results from a G to C substitution at nucleotide position 4302. The glutamic acid at codon 1434 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
