Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_017950.4(CCDC40):c.2741T>G (p.Ile914Ser), citing Ambry Variant Classification Scheme 2023: The p.I914S variant (also known as c.2741T>G), located in coding exon 17 of the CCDC40 gene, results from a T to G substitution at nucleotide position 2741. The isoleucine at codon 914 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
