Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_201596.3(CACNB2):c.841A>C (p.Met281Leu), citing Ambry Variant Classification Scheme 2023: The p.M227L variant (also known as c.679A>C), located in coding exon 7 of the CACNB2 gene, results from an A to C substitution at nucleotide position 679. The methionine at codon 227 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
