Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001128840.3(CACNA1D):c.2194T>G (p.Phe732Val), citing Ambry Variant Classification Scheme 2023: The c.2254T>G (p.F752V) alteration is located in exon 16 (coding exon 16) of the CACNA1D gene. This alteration results from a T to G substitution at nucleotide position 2254, causing the phenylalanine (F) at amino acid position 752 to be replaced by a valine (V). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be deleterious by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
