Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.2318C>G (p.Ala773Gly), citing Ambry Variant Classification Scheme 2023: The p.A773G variant (also known as c.2318C>G), located in coding exon 19 of the BUB1 gene, results from a C to G substitution at nucleotide position 2318. The alanine at codon 773 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
