Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.2773C>T (p.Leu925Phe), citing Ambry Variant Classification Scheme 2023: The p.L925F variant (also known as c.2773C>T), located in coding exon 22 of the BUB1 gene, results from a C to T substitution at nucleotide position 2773. The leucine at codon 925 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
