Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_032043.3(BRIP1):c.2210A>G (p.Glu737Gly), citing Ambry Variant Classification Scheme 2023: The p.E737G variant (also known as c.2210A>G), located in coding exon 14 of the BRIP1 gene, results from an A to G substitution at nucleotide position 2210. The glutamic acid at codon 737 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
