NM_000059.4(BRCA2):c.6422G>A (p.Gly2141Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G2141D variant (also known as c.6422G>A), located in coding exon 10 of the BRCA2 gene, results from a G to A substitution at nucleotide position 6422. The glycine at codon 2141 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
