Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000059.4(BRCA2):c.8115C>A (p.Ser2705Arg), citing Ambry Variant Classification Scheme 2023: The p.S2705R variant (also known as c.8115C>A), located in coding exon 17 of the BRCA2 gene, results from a C to A substitution at nucleotide position 8115. The serine at codon 2705 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
