Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.1969C>G (p.Gln657Glu), citing Ambry Variant Classification Scheme 2023: The p.Q657E variant (also known as c.1969C>G), located in coding exon 9 of the BRCA1 gene, results from a C to G substitution at nucleotide position 1969. The glutamine at codon 657 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
