Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000057.4(BLM):c.2216G>A (p.Gly739Asp), citing Ambry Variant Classification Scheme 2023: The p.G739D variant (also known as c.2216G>A), located in coding exon 9 of the BLM gene, results from a G to A substitution at nucleotide position 2216. The glycine at codon 739 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
