NM_000057.4(BLM):c.299A>G (p.Gln100Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q100R variant (also known as c.299A>G), located in coding exon 2 of the BLM gene, results from an A to G substitution at nucleotide position 299. The glutamine at codon 100 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
