Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004656.4(BAP1):c.932-10C>A, citing Ambry Variant Classification Scheme 2023. This variant lies in the BAP1 gene (transcript NM_004656.4) at 10 bases into the intron immediately before coding-DNA position 932, where C is replaced by A. Submitter rationale: The c.932-10C>A intronic variant results from a C to A substitution 10 nucleotides upstream from coding exon 11 in the BAP1 gene. This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis for this alteration is inconclusive. RNA studies have demonstrated that this alteration results in a splice defect; the clinical impact of this abnormal splicing is unknown at this time (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear.
