Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_024598.4(USB1):c.22G>T (p.Gly8Cys), citing Ambry Variant Classification Scheme 2023: The p.G8C variant (also known as c.22G>T), located in coding exon 1 of the USB1 gene, results from a G to T substitution at nucleotide position 22. The glycine at codon 8 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
