NM_001184.4(ATR):c.1300C>G (p.Leu434Val) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L434V variant (also known as c.1300C>G), located in coding exon 5 of the ATR gene, results from a C to G substitution at nucleotide position 1300. The leucine at codon 434 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
