Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001184.4(ATR):c.4372C>A (p.Leu1458Ile), citing Ambry Variant Classification Scheme 2023: The p.L1458I variant (also known as c.4372C>A), located in coding exon 24 of the ATR gene, results from a C to A substitution at nucleotide position 4372. The leucine at codon 1458 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
