Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001184.4(ATR):c.5298G>T (p.Trp1766Cys), citing Ambry Variant Classification Scheme 2023: The p.W1766C variant (also known as c.5298G>T), located in coding exon 31 of the ATR gene, results from a G to T substitution at nucleotide position 5298. The tryptophan at codon 1766 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
