Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001366145.2(TRPM3):c.1511T>C (p.Leu504Ser), citing Ambry Variant Classification Scheme 2023: The c.1511T>C (p.L504S) alteration is located in exon 11 (coding exon 11) of the TRPM3 gene. This alteration results from a T to C substitution at nucleotide position 1511, causing the leucine (L) at amino acid position 504 to be replaced by a serine (S). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. This missense alteration is located in a region that has a low rate of benign missense variation (Lek, 2016; Firth, 2009). This alteration is predicted to be deleterious by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
