NM_004656.4(BAP1):c.74A>G (p.Lys25Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Sema4, Sema4, citing Sema4 Curation Guidelines. This variant lies in the BAP1 gene (transcript NM_004656.4) at coding-DNA position 74, where A is replaced by G; at the protein level this means replaces lysine at residue 25 with arginine — a missense variant. Submitter rationale: The BAP1 c.74A>G (p.K25R) variant has not been reported in the literature to our knowledge. It was observed in 1/30616 chromosomes of the South Asian subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID 346126). Functional studies have not been performed, and in silico predictions of the variant's effect on protein function are inconclusive. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
