This variant lies in the TGFBR2 gene (transcript NM_003242.6) at coding-DNA position 1343 through coding-DNA position 1363, deleting 21 bases. Submitter rationale: The c.1343_1363del21 variant (also known as p.Y448_L454del) is located in coding exon 5 of the TGFBR2 gene. This variant results from an in-frame ACTCCATGGCTCTGGTGCTCT deletion at nucleotide positions 1343 to 1363. This results in the in-frame deletion of 7 amino acids at codons 448 to 454. These amino acid positions are highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
