Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003239.5(TGFB3):c.858C>A (p.His286Gln), citing Ambry Variant Classification Scheme 2023: The p.H286Q variant (also known as c.858C>A), located in coding exon 5 of the TGFB3 gene, results from a C to A substitution at nucleotide position 858. The histidine at codon 286 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
