Uncertain significance for Dyskeratosis congenita — the classification assigned by Ambry Genetics to NM_198253.3(TERT):c.2052C>A (p.Asp684Glu), citing Ambry Variant Classification Scheme 2023: The p.D684E variant (also known as c.2052C>A), located in coding exon 5 of the TERT gene, results from a C to A substitution at nucleotide position 2052. The aspartic acid at codon 684 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
