NM_000051.4(ATM):c.490T>G (p.Trp164Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W164G variant (also known as c.490T>G), located in coding exon 4 of the ATM gene, results from a T to G substitution at nucleotide position 490. The tryptophan at codon 164 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
