NM_152594.3(SPRED1):c.28A>T (p.Asn10Tyr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N10Y variant (also known as c.28A>T), located in coding exon 1 of the SPRED1 gene, results from an A to T substitution at nucleotide position 28. The asparagine at codon 10 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
