Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_003114.5(SPAG1):c.2752C>G (p.Gln918Glu), citing Ambry Variant Classification Scheme 2023: The p.Q918E variant (also known as c.2752C>G), located in coding exon 18 of the SPAG1 gene, results from a C to G substitution at nucleotide position 2752. The glutamine at codon 918 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
