NM_005633.4(SOS1):c.3095G>A (p.Ser1032Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1032N variant (also known as c.3095G>A), located in coding exon 20 of the SOS1 gene, results from a G to A substitution at nucleotide position 3095. The serine at codon 1032 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
