NM_003073.5(SMARCB1):c.64G>C (p.Asp22His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D22H variant (also known as c.64G>C), located in coding exon 1 of the SMARCB1 gene, results from a G to C substitution at nucleotide position 64. The aspartic acid at codon 22 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
