Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003072.5(SMARCA4):c.93C>A (p.Ser31Arg), citing Ambry Variant Classification Scheme 2023: The p.S31R variant (also known as c.93C>A), located in coding exon 1 of the SMARCA4 gene, results from a C to A substitution at nucleotide position 93. The serine at codon 31 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
